چکیده
AL. REINFORCES PRIOR WORK THAT SUGGESTS THAT HYPOCRETIN (HCRT, ALSO KNOWN AS OREXIN) ADMINISTRATION WILL BE AN EFFECTIVE TREATMENT FOR HUMAN NARCOLEPSY. Most human narcolepsy is caused by a loss of hypocretin neurons.1-3 That finding suggested that Hcrt administration might be an effective treatment for narcolepsy. Work in our laboratory showed that intravenous (IV) Hcrt administration reduced symptoms of canine genetic narcolepsy.4 Although the Fujiki et al. paper emphasizes technical problems associated with studying these effects, it confirms key aspects of these hopeful findings.

Systemic administration of hypocretin-1 reduces cataplexy and normalizes sleep and waking durations in narcoleptic dogs.
Recent work has implicated the hypocretin (orexin) system in the genesis of narcolepsy. In the current study we demonstrate that systemically administered hypocretin-1 (Hcrt-1) produces an increase in activity level, longer waking periods, a decrease in REM sleep without change in nonREM sleep, reduced sleep fragmentation and a dose dependent reduction in cataplexy in canine narcoleptics. Repea...

As many as 10% of humans suffer chronic sleep disturbances, yet the genetic mechanisms that regulate sleep remain essentially unknown. It is therefore crucial to develop simple and cost-effective vertebrate models to study the genetic regulation of sleep. The best characterized mammalian sleep/wake regulator is hypocretin/orexin (Hcrt), whose loss results in the sleep disorder narcolepsy and th...

The current special topics issue of Frontiers in Neuroscience “Insomnia and Beyond—Exploring the Therapeutic Potential of Orexin Receptor Antagonists” comprises 20 papers from leaders in the orexin/hypocretin field presenting their latest findings and thorough reviews, and will undoubtedly serve as an important reference for others engaged in this field of study. Since the independent identific...
متن کاملHypocretin (orexin) regulation of sleep-to-wake transitions
The hypocretin (Hcrt), also known as orexin, peptides are essential for arousal stability. Here we discuss background information about the interaction of Hcrt with other neuromodulators, including norepinephrine and acetylcholine probed with optogenetics. We conclude that Hcrt neurons integrate metabolic, circadian and limbic inputs and convey this information to a network of neuromodulators, ...

The discovery that hypocretins are involved in narcolepsy, a disorder associated with excessive daytime sleepiness, cataplexy, and unusually rapid transitions to rapid eye movement sleep, opens a new field of investigation in the area of disorders of sleep and activation. Hypocretin-1 (hcrt-1) and hypocretin-2 (hcrt-2) (also called orexin-A and orexin-B) are newly discovered neuropeptides proce...
